... from MKV (or any file) without conversion ? Why is it seemingly impossible - what's the big deal ?
The MKV would need to be BD complaint:

You can use TSmuxer to generate a BD ISO, folder, or AVCHD folder. But, like Booshman says the MKV has to be a remux and in Blu-ray spec already (not compressed or resized, etc).
